Restriction digests of the clone give the following sizes (kb): PstI--3.8; EcoRI--3.1, 0.6; PstI/EcoRI--2.6, 0.6, 0.5. Detects a TaqI polymorphism mapping to chromosome 6. A full-length Ki-ras cDNA capable of transformation if given a promoter. The PstI site on the 3' side of the insert appears to be missing. The insert contains HinfI (3), TaqI (3), Sau96I (2) and EcoRI sites. NIH 3T3 cells were transformed with DNA from the colon adenocarcinoma SW480 cell line to produce the cell line, SW2-3. DNA from SW2-3 was used to isolate the clone. |
