Restriction digests of the clone give the following sizes (kb): EcoRI/HindIII--2.9, 0.5; EcoRI--3.5; HindIII--3.5; PvuII--3.5; PvuI/PvuII--2.0, 1.5. The 5' boundary of the insert is the HindIII site at nt -374 relative to the liver-specific non-coding exon; the 3' boundary is the EcoRI site at nt +131 within that exon. The insert includes 2 regions showing homology to the glucocorticoid consensus sequence. Transcription from the T7 promoter gives an antisense riboprobe. The fragment was subcloned from the cosmid cSamD4. |
